The England right-back limped out of Tuesday night's 1-0 victory over the Midlands club and left Villa Park on crutches.
He was sent for a scan on Wednesday and although the Reds have not put a timescale on the length of his likely absence, they have confirmed the injury is to the medial ligament.
"A scan today (Wednesday) has revealed a tear in the medial ligament in Glen's right knee," a spokesperson told the club's official website.
"The player will see a specialist tomorrow after which we should be able to more accurately determine the length of his rehabilitation period."
Johnson has been one of few success stories in a torrid season for the Anfield club.
The 25-year-old has made 21 appearances and scored two goals following his £18million arrival from Portsmouth last summer.
