The Minstermen were set to stick with the same side which defeated Newport County in the FA Trophy final at the same venue last weekend.
Experienced defender Chris Doig missed that game with a calf injury and could return to a squad already lacking Scott Kerr, David McGurk and Eugen Bopp.
"Promotion is our priority but it was important to win the Trophy as that's given us confidence and proved we can win at Wembley after a couple of defeats there in the last few years," York manager Gary Mills told BBC Look North.
"The Trophy was the first the club's won and it means a lot to the club and to us. But promotion for us would be like going from the Championship to the Premier League.
"It's a tough league and I think things need to change. I don't know why only one team goes up automatically.
"Whichever way we do it, whether it be extra-time or penalties, getting the right result is the main thing."
Luton are without Godfrey Poku and longer-term casualty James Dance but striker Amari Morgan-Smith is back in contention after briefly returning for the semi-final victory against Wrexham.
