Bayern Munich travel to Spain hoping to protect their slender 2-1 lead from the first leg.
A successful night for the Germans would mean they make the Champions League final for the second time in three years.
Real Madrid boss Jose Mourinho has the luxury of selecting from a virtually full-strength squad for the semi-final second leg.
Mourinho is aware of how big the occasion is: "I don't know if the Champions League is more important than the league, because the best team always wins the league and this isn't always the case in Europe.
"However, the final of the Champions League is the most important thing in football. Today is almost like a long-awaited final, but I see calmness in the players. I don't like an excess of emotion, nor a lack of it."
Centre-back Ricardo Carvalho is a doubt with a thigh problem, while Gonzalo Higuain and Kaka are set to return to the starting line-up for the must win game.
Bayern coach Jupp Heynckes rested several players for the win at Werder Bremen on Saturday.
Bastian Schweinsteiger is set to remain in the starting XI, but Daniel Van Buyten and Breno are both out.
Heynckes said: "It is going to be spectacular. It is going to be a real treat with everything which belongs in such a football game.
"Just a fortnight ago, people were saying that Barcelona are the be all and end all, but now Real have gone and won in Barcelona.
"It will be a battle of the giants. Both clubs have a great history and I am looking forward to it, and I hope it has a positive outcome for us."
Real Madrid have a 100% home record in the Champions League this season, while Bayern Munich have won only one of their last four away games in the competition.
