The Welsh winger has been in fine form of late with eight goals and six assists in his last seven games.
However, he didn't travel with the rest of the squad to Valencia on Saturday morning as the club confirmed he was suffering from flu.
Cristiano Ronaldo also misses the match due to suspension having been sent-off in the last season's final defeat to Atletico Madrid.
Xabi Alonso has been rested as well so coach Carlo Ancelotti is expected to hand rare starts to youngsters Casemiro, Jese Rodriguez and Alvaro Morata.
