MK Dons assistant boss Karl Robinson is hoping that his side can overturn a 1-0 Johnstone's Paint southern area final first-leg deficit around on Tuesday evening at Southampton.
If they do so, he believes it could be one of the greatest achievements in the club's short history.
Two years ago they reached the final after beating Swansea on penalties at stadium:mk before overcoming Grimsby Town in the final.
However, the odds are stacked against them this time as they must travel to St Mary's and win to have any chance of making it through.
Speaking to the club's official website, Robinson said: "It's going to be a full house at a Premier League ground against a club with Premier League resources and it's a game we have to go and enjoy.
"We just hope we can come out the other side of it victorious and, if we do, it would arguably go down as the greatest victory in this club's history.
"Under the circumstances, with the injuries we've got, it's not going to be easy, but what I can guarantee is that, whoever takes to the field for this football club, the players will never let us down as individuals."
