Rovers are enjoying their best league run of the season, winning two and drawing two, but they have won just two of their 16 games against top half opponents this season.
Blackburn have lost all four of their last games against United, and have not won in the last eight. Their last victory was a 4-3 win in which David Bentley scored a hat-trick and Rio Ferdinand received his marching orders.
Blackburn are, however, a tough proposition at home this season. They have been beaten only once at home since the opening day of the season and Chelsea were held here only a couple of weeks ago.
United will be looking to bounce back after a disappointing week for the club. Last Saturday Chelsea beat them at Old Trafford to snatch the lead in the title race. Then in midweek they were dumped out of the Champions League by Bayern Munich.
Blackburn captain Ryan Nelsen returns to the squad having been out since February with a knee injury. He only makes the bench, however, with 18-year-old Phil Jones retaining his place in the starting eleven.
O'Shea, however, who replaced Rooney in midweek, returns to the starting line-up after a five month absence.
United will be without leading scorer Wayne Rooney who aggravated his ankle injury in midweek against Bayern Munich. Berbatov and Macheda start in what looks to be a more attacking 4-4-2 formation for the champions.
Manchester United team line-up: Van der Sar, Neville, Ferdinand, Vidic, O'Shea, Valencia, Scholes, Giggs, Nani, Berbatov, Macheda
Blackburn team line-up: Robinson, Salgado, Samba, Jones, Givet, Nzonzi, Grella, Emerton, Pedersen, Olsson, Kalinic

With the title race hotting up, Sir Alex Ferguson's Manchester United travel to Ewood Park looking to keep pace with leaders Chelsea.
However, with a massive midweek European game against Bayern Munich and a potential hangover from last weekend's defeat against the Blues, United will need to dig deep as they will be facing one of the top division's in-form sides on home soil.
Blackburn Rovers, who currently find themselves basking in mid-table security, have lost just three of their last 12 league games, while at home, they have suffered just one defeat in 15.
Indeed, Allardyce's well-drilled side have now taken a highly-impressive 16 points out of a possible 18 in their last six games at Ewood.
On other hand, the Old Trafford giants have won six and drawn one of their last seven top-flight clashes with the Ewood Park side.
One United player who has particularly enjoyed his meetings with Rovers is the currently injured Wayne Rooney. The striker has scored in each of his least three league games against Blackburn, although he is set to miss this clash.
Wes Brown also remains a major injury doubt although John O'Shea is back in action. Michael Owen, Anderson and Danny Welbeck remain long-term casualties, however forgotten man Owen Hargreaves is now very close to a comeback.
Blackburn will hope that a knock suffered by the returning Vince Grella in a midweek reserve match won't be enough to keep him out of Sunday's game.
Captain Ryan Nelsen also made his comeback in that second string game with Manchester City along with Yildiray Basturk. However, Irish midfielder Keith Andrews remains a major doubt as is Lars Jacobsen.
One Rovers' player who is fit and ready to play is striker Jason Roberts, who is confident his side can perform well in their last three home games, against United, Everton and Arsenal.
Manchester United were left frustrated in their hopes of stealing a march on title rivals Chelsea as they were held to a goalless draw by Blackburn Rovers at Ewood Park.

Federico Macheda lined up alongside Dimitar Berbatov in attack in place of the injured Wayne Rooney.
Blackburn had the first chance when a header from Chris Samba was collected at the foot of the left-hand post by Edwin van der Sar from a lofted through ball by Grella.
Paul Robinson in the Rovers goal was tested by a diagonal shot by Antonio Valencia following a neat one-two with Berbatov, while Gael Givet nipped in well to turn a cross from Ryan Giggs behind for a corner.
