U's maintain 100% league record
The U's, 7-1 winners at Norwich last weekend, went ahead in the ninth minute when Ashley Vincent's run ended in his shot being blocked but midfielder David Fox netted with a well-placed first-time shot from the edge of the area.
And Colchester doubled their lead, nine minutes later when Kevin Lisbie fed Simon Hackney on the left and the winger delivered a fine cross which saw Vincent headed past goalkeeper Alex McCarthy.
Vincent, Lisbie and Lee Beevers almost added to Colchester's lead before the break and Yeovil failed to trouble U's goalkeeper Ben Williams in the first half.
Lisbie almost added a third when through on goal but his touch let him down and McCarthy saved well from Vincent.
Ryan Mason pulled one back with a fine curling free-kick with eight minutes to go and Yeovil pushed for an equaliser, but Colchester held on for victory.
