Roy McFarland has ended his four-year association with Chesterfield after the 1-0 home defeat to Leyton Orient left the Spireites only one point above the League One relegation zone.
Lee Richardson has been appointed caretaker manager at Saltergate until the end of the season.
Chairman Barrie Hubbard told the club's official website: "During my long involvement with the club I've made a number of mistakes but appointing Roy McFarland as manager wasn't one of them.
"He's helped return respect from the football world to a club recovering from the shenanigans of a previous regime and we'll always thank him for that.
"Lee Richardson has been a loyal assistant to Roy McFarland and has worked hard to achieve relevant qualifications and now deserves the chance to take over the reins until the end of the season."
McFarland was appointed manager in May 2003 having previously being in charge of Bradford City, Derby County, Bolton Wanderers, Cambridge United and Torquay United.
However, a run of seven defeats in their last ten matches led to the club's decision to part company with McFarland.
