Canada international Hume, who sustained the injury after being elbowed by Sheffield United skipper Chris Morgan at Oakwell last November, followed up a 45-minute run out last week at Gainsborough by completing 90 minutes in a 2-0 win over a Hull XI.
Tykes boss Simon Davey told the club's official website: "It was a big step for Iain to get through 90 minutes and another hurdle to overcome on his way back to fitness.
"It will have done him the world of good to play the full game. He just needs a few more to get that match fitness and sharpness back."
Goals in each half from summer signing Onome Sodje and Michael Coulson, on his way back from a second cruciate knee ligament injury, secured the Tykes a 2-0 win at Oakwell.
